It's not canon, and here's the definitive proof. Ready for the ride?
In the video game, Egon has a nephew named Ed.

Ed Spengler, nephew of Egon, is a character in fan films Freddy vs Ghostbusters and Return of the Ghostbusters.

So far we proved that the game and those fan films are set in the same universe, but the rabbit hole goes deeper.
The Angry Video Game Nerd (meaning the character played by James Rolfe, not James Rolfe himself) exists in the same universe as he has various brief scenes in Return of the Ghostbusters.

In the Toxic Crusaders episode, the AVGN meets Lloyd Kaufman, who makes a peculiar remark: "But you folks out there in Videoland...", implying outside the AVGN's house is a place called Videoland.

In the Street Fighter 2010 episode, we see the outside of the AVGN's house. It's made up of 2D clipart and slightly blurred sprites from old video games. So this is what Videoland, whatever it is, looks like.

In 2007, the AVGN episodes were released on a website called Screwattack. The same website also hosted another web series: The New Adventures of Captain S, about a Sega Genesis enthusiast who enters a parallel world called Videoland via a magic cartridge. The parts of Videoland we see are made up of 2D clipart and slightly blurred sprites from old video games, and the Head (the leader of Videoland) states that it's comprised of all video games.
Every single video game is part of Videoland, remember that.
The Captain S webseries also has a special crossover episode with the Angry Video Game Nerd, which, while stated to be non-canonical for the Captain S series, is canonical to the AVGN series, because a regular AVGN episode is a direct continuation of the crossover. Again, we are shown the AVGN in Videoland, with the proof that the Videoland in the Captain S series is the same Videoland in the AVGN series.

So, if the AVGN series is set in Videoland, then so are the two Denver Ghostbusters fan films, and so is the video game... which makes sense, because Videoland includes all video games, including the Ghostbusters game!
